●  研究兴趣、领域:


高分辨冷冻电镜断层成像(cryo-Electron Tomography, cryo-ET)是将置于透射电镜样品台上的冷冻样品沿固定轴旋转,并从不同角度拍照获得倾转系列(tilt series, TS),将其重构为三维断层图像,再使用子断层平均法(sub-tomogram averaging, STA)进行结构解析的新型结构生物学方法。由于cryo-ET原始数据包含精确的Z轴信息,它对样品纯度及构象均一性要求较低,且在制备样品过程中无需使其脱离原生环境,获得的结构更具生理意义,擅长提供的结构信息有:1)蛋白在其载体上或功能状态中的原位结构;2)大分子复合物的高级结构,包括蛋白组装、蛋白/脂膜间互作的landscape;3)不易在体外重组表达的复合物结构。
本课题组的研究兴趣包括:1)cryo-ET方法开发:高通量、亚纳米分辨率cryo-ET方法;光学-电子显微镜联合使用(CLEM)及聚焦离子束(FIB-SEM)减薄细胞及原位结构;2)新发致病型囊膜病毒的组装及通过膜融合去组装的结构与功能。
