● 研究兴趣、领域:
1. 高分辨冷冻电子断层术(cryo-ET)。高分辨cryo-ET是使用冷冻电镜收集三维断层图像,再使用子断层图像平均法进行结构解析的跨尺度原位结构生物学方法。Cryo-ET不仅擅长解析动态组装的巨型分子机器结构,与结合光电联合成像(CLEM)、冷冻聚焦离子束(cryo-FIB)等技术结合,更可在细胞中进行成像和大分子结构原位解析,获得具有生理意义的原位结构。Cryo-ET技术发展迅速,正填补从病毒到细胞,甚至组织维度间的结构生物学空白。
2. 新发囊膜病毒的原位结构与功能。我们以cryo-ET为主要方法,系统研究多种新发致病型囊膜病毒组装、感染、及免疫的原位结构及机制,最终目的是找到病毒的弱点,并为抗体、疫苗设计提供新思路。
