我们了解生物机体的一个主要的障碍是其在组织水平上的复杂性。基膜(basement membranes,BMs)是由高度保守的细胞外基质蛋白的聚合物,它与其组分为所有动物组织都提供了必要的结构性支持。在细胞的形态发生,细胞信号,免疫反应,细胞再生和肿瘤形成的过程中,基膜与其组分都起着关键的作用。我们在最近的研究中发现,基膜的两个重要的组成成分IV型胶原蛋白(Collagen IV)和串珠素(Perlecan),发挥相反的作用力来决定细胞和器官的形状。目前本实验室正在进行的几个项目,研究的就是基底膜各组分如何分泌并组装成多聚体及其在发育、再生和癌症中的作用。
